Building on our earlier work defining the signaling pathways governing bovine trophoblast stem cell (TSC) self-renewal and differentiation, we generated transformed bovine TSC (bTSC) lines. These cells not only maintain their stem-like identity under ROCK inhibition but also undergo appropriate trophoblast maturation in response to TGFβ. Importantly, the lines are highly amenable to genetic manipulation—supporting stable lentiviral gene introduction, CRISPR/Cas9 editing, and reporter expression—making them well suited for functional studies.
